If you're even considering the Lexus, then you might prefer the LX and it's climate control. After getting spoiled by a Volvo and an Acura RL, I would have liked to be able to set a temp and forget it. I can't however, speak to how successful the Santa Fe pulls of the auto-climate control.
If you stick with the GLS, I recommend getting one with ABS and Traction Control System (TCS) added.
As for 2WD vs. 4WD, I got the 4WD without doing my homework. The 2WD is FWD and would have handled 99.99% of my driving needs. The 2WD owners say they are a little zippier (is that a word?).
I'm still very impressed with our Santa Fe and the wife especially likes it.
